The nRF51 does not have USB, so you cannot program it over USB directly. You need some kind of external programmer for the initial programming. E.g. the onboard interface chip on the DK, or a SEGGER J-Link probe or something else. For later upgrades you can use DFU, if you have programmed the device with a bootloader.
