&lt;p&gt;Yes, that is possible, but you need to change the VDD voltage of the nRF5340DK to 1.8V. When the USB connector is attached the VDD voltage will be regulated to 3V by default, if you supply 1.8V to the VIO_REF connector you can change the output of the regulator to match your custom board voltage.&lt;/p&gt;
&lt;p&gt;Another option is to just supply the custom board with 3V from the DK when programming/debugging if your board supports having a higher VDD voltage.&lt;/p&gt;
