In the BSP startup files for the nRF5340 (gcc_startup_nrf5340_application.S & gcc_startup_nrf5340_network.S) the interrupt vectors USPTn_IRQHandler (n=0,...,3) are defined.
You can use any free General purpose I/O for SERIAL. https://infocenter.nordicsemi.com/topic/ps_nrf5340/chapters/pin.html?cp=4_0_0_8_0

For the nRF53DK, we use this pin mapping: https://infocenter.nordicsemi.com/topic/ug_nrf5340_dk/UG/dk/vir_com_port.html

I'm not seeing this defined in the latest MDK files. https://www.nordicsemi.com/Products/Development-tools/nRF-MDK/Download?lang=en#infotabs , if you are using very some old files, then there might have been a bug with that file.
