&lt;p&gt;&amp;nbsp; Yes, the issue has been resolved now. Reason for this issue is that Internal 32 kHz clock is not initialized. After adding the line &lt;span style="background-color:#f8f8f8;color:#1d1c1d;float:none;font-family:Slack-Lato, Slack-Fractions, appleLogo, sans-serif;font-size:15px;font-style:normal;font-weight:400;letter-spacing:normal;text-align:left;text-indent:0px;text-transform:none;white-space:normal;"&gt;CONFIG_CLOCK_CONTROL_NRF_K32SRC_RC=y&lt;/span&gt; to our build, it is working fine.&lt;br /&gt;Now we can able to run the boot in our custom boards and it is working.&lt;/p&gt;

&lt;p&gt;Actually we done the boot build for our custom board and flashing the file in our board via Seggar.&lt;/p&gt;
&lt;p&gt;After flashing, we are checking the log messages in UART1(P1.00 &amp;amp; P1.01) lines. In that we are getting the above message.&lt;/p&gt;
&lt;p&gt;Also, we checked for log messages in other UART2, no messages were found. The thing, we would like to know that we done the Build configuration correctly or not?&lt;/p&gt;

&lt;p&gt;Sorry, I was sick, which caused a delay in responding.&lt;/p&gt;
&lt;p&gt;Let&amp;#39;s clarify a few things before moving further. I assume you are trying to flash your first programme to see whether the newly developed hardware is working fine. For flashing your first application, you are trying to put the custom board in the bootloader mode. But have you flashed the bootloader to your custom board? (I assumed you had done this and responded; sorry for that.) If not, you need to flash the bootloader (MCUboot). You can use J-Link for this. Build mcuboot for your custom board and flash it using the command-line tool.&lt;/p&gt;
&lt;p&gt;To enter bootloader mode, you need to trigger the bootloader on your board. For triggering, have you configured it just like what is done in DK?&lt;/p&gt;
&lt;p&gt;If your board enters bootloder mode, then you can generate a DFU package using nrfutil and flash it over UART. Read this &lt;a href="https://devzone.nordicsemi.com/nordic/nordic-blog/b/blog/posts/nrf-util-unified-command-line-utility"&gt;blog on nRFutil&lt;/a&gt;&amp;nbsp;for generating and flashing the DFU package.&lt;/p&gt;

&lt;p&gt;AFAIK, this is enough to make the 5340 enter bootloader mode. Are you using a USB-UART adapter for flashing your custom 5340? Could you share some more details on the connection? Have you tried communicating to your board from a terminal (like by using nrfutil or by using a serial terminal like PuTTy)? Have you opened the device inside the &lt;a href="https://www.nordicsemi.com/Products/Development-tools/nRF-Connect-for-Desktop"&gt;nRF connect for desktop App&lt;/a&gt;? If so, what is the response from the device?&lt;/p&gt;

But we couldn&amp;#39;t do it, need instruction for the DFU in our device.&lt;br /&gt;We got our SCH &amp;amp; PCB layout verified by team, now we completed the SMD and now started testing our device. We are trying to flash our device over UART. Device is turned ON, as I can check the power lines and it is same as in the EVK, but not able to flash.&lt;/p&gt;
