Thanks. I am wondering if there is any other way to connect this board even using the nRF52840DK board. I don't have a USB port on this custom board, and there is no way to include one now.

Hello Gautam,

You have to connect board with PC through USB port. The USB cable is necessary for both power supply and data transfer. After flashing application on board, you can connect to any central device through BLE. That's the further step but it is mandatory to have USB port on board.

For programming a custom board, you will need a debug unit too. So you can use this to connect to the PC. Please take a look at the following link:
