Hi,

As shown in the key features, nRF5340 audio DK has mono output and stereo line input. It is possible to connect external hardware codec. You can get more information about needed application changes in this ticket. For receiving stereo signal and hardware connections you can refer to this ticket.

Looked at the codec chip itself? It only has a mono amplifier output.

In order to properly connect stereo headphones you need a different codec chip. It may be possible to connect another codec onto the AUX I²S interface.

Firmware for that will be your task though.
