I take this code refernce and manually drive the pin it working now Modbus through UART

Then you should also check the config file(s) in the build dir. There is a surprisingly large amount of peripherials enabled by default.

Im using Only UART for MODBUS in my project it is only enable (Ble+Uart Only), other peripheral are disabled so there not be conflict of Pins, might be Modbus driver have issue!

Check the DTS files in the build dir, the pins may be configured for other peripherials.
