Did the application that you updated with DFU have DFU support, i.e. does it have the DFU Service? If not then you need to add DFU support to the application as explained on this Infocenter page.

I found a major problem now. Once I have updated with the image file, when I tired to load the same file again it shows the same error DFU service not found. Can you advice on this please.

It seems that I can only update for one time.

Is your nRF52 device advertising as DFUTarg? (See this infocenter page for a step-by-step guide on running the bootloader example)

Have you flashed the bootloader, softdevice and an application that has DFU support to you nRF52 device?

When I am trying to load ".Zip" file, it says "Upload Failed: DFU service not found". Am I missing Something here?

You need to create a firmware image zip as explained here.
