It could be an issue with the HWFC. You could try to disable it in the serial bootloader project and in the nrfutil source as described in this answer.

I am using a NRF52 DK yes, connected via USB to my UP Board

By dev board you mean the nRF52 DK? Are you able to provide a logic trace of the of the UART line between the UpBoard and the nRF52`?

I am having issues with both Serial and BLE on the UpBoard, i put some logs in the dfu_transport_serial.py and it looks like it is failing on the CRC validation, the CRC from the response is incorrect. i don't know why this would be happening? do you have any ideas, i used the same package on a Virtual Machine Ubuntu and it works fine. i am currently using a dev board connected via USB

Are you only seeing issues with the serial bootloader on the UpBoard or both BLE and serial? How are you connecting the nRF52 device to the UpBoard, just TX and RX or TX, RX, CTS and RTS?
