No, you should only need to flash it once. But if you accidently erase it somehow, you would of course need to flash it again.

Try nrfgo studio, and see if the softdevice is on the chip

Did you flash the softdevice hex? its located in sdkfolder\components\softdevice\s140\hex

But for example "blinky" everthing is good. Only for examples from ble_peripherial I have problems.

Use nRFGo Studio if you want a GUI: www.nordicsemi.com/.../nRFgo-Studio

Commandline: use nrfjprog

Sounds like you have forgot to flash the S140 softdevice. You have only built and flashed/loaded the application, but you also need to flash the S140 softdevice(the BLE stack) to the chip
