Yes, with nRFConnect you can check what services the device has. You dont need a DFU packet to check that. If the device has the DFU BLE service, it should look something liks this:

The DFU zip file is necessary when test DFU function if I use nRF-Connect, to do this job? Can I test DFU function available without DFU zip file?

It faster to test with nRF-Connect, but yes, you can use nRFToolbox also. You will need a DFU zip image to test with nRFToolbox. If the device does not have the DFU characteristics, nRFToolbox will report back that DFU Failed: DFU characteristics not found.

You can try to make the device enter DFU mode, either by holding one of the buttons on the device down during startup, or by connecting to it using nRF Connect for mobile, and see if the device has the DFU control point characteristic.
