a. Don't use the template example it's too bloated, use BLE Secure DFU Bootloader instead.
b. You can use more than 256kB if you're using 'Single-bank DFU'. See Dual-bank and single-bank updates.
You can use any peripheral in SystemON mode.
You can add the driver/libraries you need to the example given in (1.). There are some peripherals that are used by the SoftDevice and is therefore unavailable to the application. See System on Chip resource requirements.
Another alternative is the ble_app_uart application.

1) How about Image up-gradation or DFU feature is supported in all BLE Peripheral examples.

and my previous queries.

For time being I am planning to use “System ON” low power mode. Whether can I use RTC timer for this. Any reference for using RTC as “System ON” mode ?
For I2C & SPI interface do I need to add files from Peripheral example code. My worry is difference between SoftDevice Enabled/Disable example code. Whether can I add those files directly into 'ble_app_template' project.

If you're taking that from SES, I think it includes the SoftDevice.

Yes, the Template does include a load of stuff like bonding, peer manager, etc - which you could do without.

Look at the other peripheral examples, and see which comes close to your requirements.

In particular, the Blinky & Nordic UART ...
