There is usually no communication between the nRF9160 and nRF52840 on the nRF91DK. The nRF52840 is used to control a set of switches that routes some of the nRF9160 GPIO pins.

However, some pins are connected between the nRF9160 and nRF52840. These are not normally used but can be used to set up communication between the two chips. One sample that does so is the lte_ble_gateway, which uses the nRF52840 as a network processor, running BLE HCI over UART.
