Softdevice is not needed.
For some reasons now I am seeing the same issue as yours. I checked with the developer and the issue is that the NRF Connect DTM application couldn't detect the PCA10100 board, but it also didn't take the board as custom board, so it throw the error. We will add support for it in the next release of nRFConnect DTM.

I tried your hex file and the result is the same.

Do i need to flash anything else? (SoftDevice?)

Could you capture a screenshot of nRF Connect when you see the issue ?

I just tried to test here and it worked for me.
Could you try to test using the attached hex file ? Please use nrfjprog to program and do a reset after you program it.
