There has been a lot of changes to the DFU functionality recently, so you should try to use most recent versions of all the components.

What versions of nRFgo Studio, nRF51 SDK and the smartphone app are you using? If you are using the most recent versions of all of them, at it still does not work, I am not able to help you I'm afraid.

I flash the Softdevice and Bootloader using nRFgo Studio. and Download the application using Keil uVision4.
I flash the Softdevice and Bootloader using nRFgo Studio. then I use a smartphone app to upload an application to the device via DFU.

Case 1 is no operation. and Case 2 is operation. but if I re-upload application to the device via DFU, No operation.

I am not sure I understand your question, but I will give it a go.

The bootloader is capable of flashing hex files to your device in the same way as conventional wired options (J-LINK over USB etc.)

The typical use case would be that the device comes shipped with all three hex files in place (bootloader, softdevice, application), and that when an update to one of these hex files is required you would use the bootloader to perform this update.

This way you can flash your device without having to connect a wire to it.
