I'm afraid that there's no step-by-step guide anywhere, but you should browse this forum as there are many questions and answers about the topic. There is also an application note called nAN-34 Setting up production test using DTM.

The "Set up on" option in nRFgo Studio is intended for other kits and should not be used for the nRF51 DK. Just chose the correct COM port instead. Remember to program the DTM example code from the SDK as well as the corresponding softdevice onto your DK.
