It is true that the USB device is limited to 12MBps, and the actual data throughput is even lower.

The SPIM3 peripheral should achieve close to 32Mbps when running at the max 32MHz frequency.

The QSPI goes higher still, up to almost 128Mbps, but that doesn't really help if you are looking for increased USB throughput.

Nope, the USB device only works with 12 MBps full speed.

On the NRF52840 I cannot see any peripherial that can sustain 27MBps throughput. You will need a µC with more "beef".
