&lt;p&gt;Unfortunately, the chip is powered down in System OFF mode. This means that resources such as clocks and timers will not be available. Please reffer to the &lt;span&gt;&lt;a title="System OFF mode" href="https://infocenter.nordicsemi.com/topic/ps_nrf5340/chapters/pmu/doc/pmu.html?cp=3_0_0_3_1#unique_1492065360"&gt;System OFF mode&lt;/a&gt;&lt;/span&gt; section of the PS for more details.&lt;/p&gt;
