Hi,

Are you using nRF52840DK? If so, You can use the GPIOs on the Connector interface, but avoid some dedicated pins as the note on the Connector interface doc.

Have a look at the SDK TWI examples, the pins of SDL and SDA are defined in pca10056.h under nRF5_SDK_17.1.0\components\boards.
