Ok. Some of the GPIOs on the nRF9160-DK are used for LEDS, buttons, etc. Here is the list over used GPIOs: https://infocenter.nordicsemi.com/topic/ug_nrf91_dk/UG/nrf91_DK/if_connector.html
I recommend using GPIOs that are not in that list when connecting to the ENC424J600.

Right now I don't have one. But I expect to buy one and test soon as possible.

Yes, but do you have any boards/shield with ENC424J600 now? If yes, what is the name of the ENC424J600 board/shield?

Hi,

Yes, that should be possible, as this ENC424J600 can interface with the nRF9160 using SPI. The easiest way to test this is if you have some small board or shield with the ENC424J600, that you can connect with some cables to some of the available GPIOs on the nRF9160DK. Zephyr already have drivers for this ethernet device: github.com/.../eth_enc424j600.c
