Please refer to the CMW270 documentation for how to connect UART over USB. Also remember to set 2-wire UART and not HCI.

You can use any free GPIOs as UART pins. You will need an external TTL to RS232 converter in this case.

Thanks for replying. I connect nRF Connect for Desktop to nRF5340 DK via USB.

I've tried the same way to let CMW270 control nRF5340 DK, but CMW270 cannot detect nRF5340 DK.

Should I replace USB with UART?

If so, which GPIO should I use?

If it works with nRFConnect for desktop, then the DK with the DTM FW should be ok. The CMW270 should be able to communicate on the same UART as the PC does, with nRFConnect for desktop.
