&lt;p&gt;If you configure nrf_serial to use UARTE1, there should not be any compatibility issues with the OpenThread stack that uses UART0, just make sure that you disable UART0 for the UART driver in your sdk_config.h file (UART0_ENABLED and&amp;nbsp;NRFX_UART0_ENABLED set to 0).&lt;/p&gt;
&lt;p&gt;I&amp;#39;m not sure where the conflict with RTC2 is coming from, as nrf_serial should use app_timer (RTC1) for timeouts. Can you check the full build output, which should tell you both locations of the conflict?&lt;/p&gt;
&lt;p&gt;That said, I would recommend you to look into using &lt;a href="https://infocenter.nordicsemi.com/topic/sdk_nrf5_v16.0.0/lib_libuarte.html"&gt;libUARTE&lt;/a&gt; instead of nrf_serial. nrf_serial is deprecated and removed in nRF5 SDK v17.x.x, and is generally not as mature and feature complete as libUARTE.&lt;/p&gt;
