I'm looking to create a custom board with MDBT50Q-1MV2. Can I directly power it from a LiPo battery (3.7V - 200mAh) only to the VDDH pin? My board is very compact, so I want to simplify as much as possible. Can you guys give me some advise ?

You can power directly through VDDH. The nRF52840 has internal regulator that you can set the operating voltage you want. Powered this way the VDD_nRF will be output based on the settings. Current will be limited so you cannot put to much load on it.

Yes, that is true. External circuitry can then be powered through VDD.
