Can this SIM_DET pin detect the SIM card at this stage?

SIM_DET pin is not used. Should be left floating to reduce current leakage. 

If you need to check if there is a SIM card in the socket you can use specific AT commands for that. Like https://infocenter.nordicsemi.com/topic/ref_at_commands/REF/at_commands/access_uicc/xsim.html?cp=2_2_0_12_0 .
