When will the power profiler be updated to support the larger LL payloads (251 bytes)?

Yes, that's in my schedule. I can't give you an estimate quite yet on when this will be added though. Meanwhile, you should get a pretty good estimate on current consumption by extending the radio TX period with the time it takes to transmit the extra bytes. I.e. 250 bytes @1Mbps = 2000µs.
