Is it OK to disconnect or short the load of nRF52 RF port ? Can the TX withstand the reflected back signal at maximal transmission power ?

That will not be a problem. The absolute maximum ratings in the RF input level for the nrf52832dk & nrf52840 preview dk are 10 dBm (see link). Even if you set transmission power to max, the reflected signal will never reach 10 dBm. Hope that answers your question!
