  I used nRF52840 dongle device to run BLE multi-link Central and nrf52832 to run BLE Blinky example to test the communication of 20 nodes. How can I set and verify that my wireless speed is 2M, and how can I set the long packet mode (maximum 251bytes).I want to achieve a large number of data communication, bluetooth parameters of this area to pay attention to what?

To make sure that you have 2Mbps enabled, check that primary_phy is set to BLE_GAP_PHY_2MBPS in your main.c file. 
To enable long packet mode, have a look at this case.
