Wireless range depends on a lot of factors, but 5-6m sounds quite short. You can make a private case and share your design if you want us to review it.

We dont find design file for this module.

Well, 8-9m when connecting to your phone is not a whole lot, so if you have similar poor performance in both ends of the link, further reduction is expected. What range do you get if you run the same application on a DK?

Based on this. Will i be able to communication in ble@1Mbs mode between two nrf52840-dk module at a distance of 50 meters?

It depends on the environment you are testing in. If fairly noise free and line of sight then yes, if in a dense office building in a busy city then probably no.

I tested two nrf52840 Holyiot-18010 module.
The central-link was in the build and the peripheral was in the yard. I could communication between two board at a distance of 20 meters. I think is very good performance! I think there was something wrong with the PTR9818 module.

I do not think there is anything wrong with the module, but the antennas are likely affected differently if you do not mount the modules on a host PCB, as they are optimized for and designed for this. Also, the antennas are likely affected differently if you do not follow the layout/mounting requirements from the manufacturer.
Still, there is only one ground pin on the PTR9818 it seems, meaning it will not make for a very good ground plane connection with the host PCB. This will likely limit RF range also.

You were right. PTR module work nice. I placed the module an other 230x230 mm PCB and I soldered the gnd to the pcb. I measured between two module 150m distance on BLE 4.0 mode.
So i had mistake on my pcb design.
