Probably not, based on this snip from the product specification "nRF52840_PS_v1.1.pdf"..?

Hi, this is not what I mean. I don't like to use D+/D- as GPIO pins. In the layout, I would like to use AC9 but route the trace from AC9 via AD6.

My initial thought was also to reply "Probably not". I have now checked with other team members who say it is not recommended. VBUS, D+ and D- belongs to a different power domain than the GPIOs and also have internal hardwired circuitry.
