I would like to check if it is possible to setup 3 sets of nRF5340 Audio DK as a master with two slaves.
Where the master and salves are able to send audio (via mic) in both directions?

Do you mean set as the gateway, the left headset, or the right headset as in the Connected Isochronous Stream (CIS) mode? If so, CIS is a bidirectional communication protocol, but in the current version of the nRF5340 Audio application, the CIS mode offers only monodirectional communication. See this Application modes doc.

Yes, the CIS mode seems more appropriate to what we have in mind
Does monodirectional mean that when i sent audio from the headset MIC to gateway. The data from gateway will be disrupted
or
the Monodirectional means data can only be sent from gateway to headset?
