Could we flash an arduino nano 33 iot with nrf52840 dk to use zephyr?

Hello Ghazi,

The arduino nano 33 iot is supported here: https://developer.nordicsemi.com/nRF_Connect_SDK/doc/latest/zephyr/boards/index.html. So yes, you can use an nRF52840-DK as well as any other JLINK  to program the board.
