1.CIS mode,when is bidirectional communication supported?

2.when LC3 support 16bit 16kHZ?

3.when suppourt DFU OTA?

1.CIS mode,when is bidirectional communication supported?

Bi-directional communication is supported in CIS mode. You can refer this portion in our developer page. Please refer to this ticket too.

2.when LC3 support 16bit 16kHZ?

LC3 does support these. Please refer this.

3.when suppourt DFU OTA?

This support was added in nRF Connect SDK v2.1 onwards. Please refer our documentation.
